In order to get my new software running on my desktop, I want to add some extra RAM. At the moment it only has 2x256. According to a crucial scan I could put 2 x 1G, but it also seems to suggest I can put in 1x1GB and keep the other 256. Is this right? I only really want to get to !GB to make sure this software works.

If you're upgrading to 1GB it's probably cheaper to buy 1x 1GB stick than 2x 512MB sticks - and considerably cheaper if in the future you want to increase your capacity again, as you'd only have to buy another 1x 1GB to reach 2GB, and not 2x 1GB. You'll also end up with 1.25 GB rather than 1GB doing it their way.

Sorry, no they do suggest buying 1x1GB and keeping the 256MB in, not getting 2x512MB. I just seem to remember being told once that you had to have the same in both slots. Just wanted to check before I ordered it.

Not really. If you have 2 different speeds of RAM in then the slowest speed (not talking about the capacity of the RAM here) is what you'll get. But you can mix and match so long as it's all compatible with your PC, and you can leave slots empty if you didn't have enough sticks to fill them, though it's better to fill them all if you can.
